Butt and Guthrie will definitely be back in contention according to Magpies boss Joe Kinnear despite the former missing last week's 2-2 draw with Stoke City and the latter limping off in the first half.
Republic of Ireland winger Duff has missed the last two matches with a groin problem, but has an outside chance of featuring at Fratton Park.
"It wasn't ideal against Stoke on Saturday in midfield because we'd already lost Nicky and Damien before the game and then Danny got injured in the first half," Kinnear told the club's official website.
"But looking ahead to Sunday, we'll probably have Butt and Guthrie back and maybe, just maybe, Duff as well."
